This kit utilises our HOOK™ Activated Agarose (Amine) that has reactive aldehyde groups. The aldehyde groups of the agarose react spontaneously with primary amines, located at the N-terminus of proteins or in lysine residues, to form intermediate Schiff Base complexes. These, in turn, are selectively reduced by the supplied reduction agent. Following quenching of the reaction the protein or peptide is linked to the agarose by stable amine linkage. The resin can now be aliquoted into the supplied columns for use in purification and interation studies.
In addition to all the coupling reagents, the HOOK™ Agarose Coupling Kit is supplied with a specific Optimizer Buffer™ that provides the optimal coupling conditions and can be used to equilibrate your starting protein sample.
This kit is suitable for the preparation of 5 stable columns containing 2 ml resin.
